Do Thomas and Brenda end up together in the Maze Runner?
Brenda is a character who is first introduced in
Thomas (Love Interest/Savior) Teresa Agnes (deceased) Newt (deceased) Minho.
Thomas whines about missing Teresa, but not Minho or Newt even though Thomas has spent more time with those two than he ever had with Teresa. Even in the first book, he has a few interactions with Teresa at most before the other Gladers are already making comments about Teresa being Thomas' girlfriend.
The last shot is of him eyeing the boat they came on, holding the cure in his hand. Thomas survived the Maze, he survived the Scorch, and even helped destroy WCKD. To the delight of many fans (especially all the Brenda-Thomas shippers out there) Thomas is now free to live his life in peace with his friends by his side.
Thomas and Teresa share several kisses during the story. Brenda kisses Thomas' cheek. She pulls his head toward her to try and kiss his lips, but he pulls away. Aris and Teresa share several kisses, which spark Thomas' jealousy.
Brenda has a crush on Thomas and flirts with him consistently. However their relationship is soured after drugs cause her to act on her feelings and him to insult her. Rat Man: The first WICKED researcher to identify himself to the Gladers. He informs them that they are infected with the Flare and how to get its cure.
Either way, she sacrifices herself for him. In the film, Thomas kisses Teresa. In the book, he kisses Brenda for the first time once they reach the Safe Haven.

single. But Thomas does form a close friendship with Teresa, the only girl to ever arrive in The Glade. Though he can't really remember details of his former life, Thomas has been dreaming of Teresa's face even before she arrives.

Why is Thomas's blood the cure?
In the movies, no. However, in the books his blood is not the cure to the Flare, it is merely that he is the “prime candidate” and that his brain has provided the best data that will be most helpful in creating a cure.
Thomas tells the Keepers that he and Teresa were manipulated into designing the Maze because they share the gift of telepathy. He convinces the others that he and Teresa are now in as much danger as them, but he knows the way out. Thomas tells them in order to escape, they have to jump through the Griever Hole.
The Maze Runner movie series has ended, but James Dashner has released prequels that would make future movies unlikely. The fourth and fifth Dashner novels focus on new characters and take place before the events of the Maze.
Brenda is infected with the Flare virus, but one of the members of the Right Arm, a former WCKD scientist, uses an enzyme cure that fully heals Brenda. The scientist reveals to Thomas that the cure can only be harvested from an Immune's body, not manufactured.
While the much-delayed Death Cure film offered a more romantic goodbye, it is highly probable that “the only thing Teresa ever cared for” might be Thomas himself. Given her intentions of saving the Gladers and fighting the virus, she might have cared for humanity's survival too.
The author never states that any of the characters are explicitly Lgbtq+. There isn't much romance anyway though, and only two real couples. The reason you may be confused is many people do believe that the main character (Thomas) is bi.
They move in to kiss, but Thomas only sees himself kissing Teresa. He tells Brenda she isn't Teresa, leaving her disappointed. After experiencing trippy visions, Thomas wakes up to find Teresa and Minho sitting over him. He and Brenda are back with Jorge and the Gladers, and Thomas finds Jorge beating Marcus's face in.
Fire Juice is the mixture of drugs and other unknown materials/ingredients that Thomas and Brenda drink at the Crank Party in The Scorch Trials. It tasted like fire and eventually made them pass out. In the movies, it was given to them by Marcus, who they didn't know at the time.
This scene happens later in the film, but when Brenda tries to kiss Thomas in the book, he refuses because of his feelings for Teresa. He doesn't refuse in the film, but he does tell Brenda that “You're not her”, which she doesn't appreciate. Still, they are not supposed to kiss until the third book.
In the books, Newt got infected at some point in The Scorch Trials, most likely as they made their way through the city and were exposed to the Flare for the first time. So in the beginning of The Death Cure, he would have already had it for about a month, which is why he got worse so quickly.
Was Brenda's brother in the maze?
In the Scorch Trials film, Brenda carries a silver trinket box around with a boy's photo in the lid. She eventually tells Thomas that the boy was George, the same George that died in the Maze.
