Formaldehyde stock solution (20%) (2024)

Paraformaldehyde, EM grade

NaOH (1 N)

HCl (1 N)

Add 200 mg of EM-grade paraformaldehyde per milliliter of H2O. Heat at 60°C on a stir plate in a ventilated chemical fume hood to dissolve. Add a trace of NaOH to help dissolve the paraformaldehyde (no more than 1 mL of 1 N NaOH to 100 mL of H2O). Do not overheat; if the solution boils or turns brown, discard and start over. Adjust pH to ~7.0 with 1 N HCl. Formaldehyde is typically diluted to 4% in the desired buffer.

How do you make 20% formaldehyde? ›

To prepare a 10% solution, add 1 volume of formalin to 9 volumes of water; for a 20% solution, add 2 volumes of formalin to 8 volumes of water. The 10% solution contains approximately 4.1 – 4.5% formaldehyde; the 20% solution contains approximately 8 – 9% formaldehyde.

What is the stock solution of formaldehyde? ›

Preparation of a Stock Solution

In a calibrated or conformity-checked 1000-mL volumetric flask make up 2.50 mL of formaldehyde solution min. 37 % to the mark with distilled water. The stock solution prepared according to this procedure has a concentration of approximately 1000 mg/l formaldehyde.

How to make 20% PFA? ›

Paraformaldehyde (20%)

Prepare 20% paraformaldehyde by boiling distilled water and then adding 20 g of solid paraformaldehyde per 100 mL of H2O with stirring. Add NaOH pellets until the paraformaldehyde is dissolved. Aliquot in 50-mL tubes and store at −20°C.

How do you make a 2% formaldehyde solution? ›

Add 2 g paraformaldehyde powder (e.g., Sigma, St. Louis, MO) to 100 ml of 1 X PBS. Heat to 70°C (do not exceed this temperature) in a fume hood until the paraformaldehyde goes into solution (note that this happens quickly as soon as the suspension reaches 70°C). Allow the solution to cool to room temperature.

How to make 10% formaldehyde? ›

10% formalin is a 1:10 dilution of 100% formalin in water, i.e. 1 part saturated formalde- hyde in water diluted with 9 parts plain water. Since 100% formalin contains 40% formaldehyde, a 1:10 dilution would contain 4% formaldehyde.

Why is formaldehyde banned? ›

Formaldehyde, Cosmetics, and Other Countries

The FDA is not the first regulatory body to consider banning formaldehyde in cosmetics. On July 8, 2022, the European Union banned formaldehyde-containing cosmetics after formaldehyde was officially named a carcinogen, known as a cancer-causing chemical.

What is 40% formaldehyde solution used for? ›

Formalin 40 is widely used as a disinfectant in poultry houses and hatcheries. It is also used in fisheries as a parasiticide. It can be used as a general disinfectant. Soaking for 30 minutes with a 10% solution of Formalin 40 will destroy a wide range of disease causing bacteria and viruses.

What is formaldehyde 10% solution used for? ›

Formaldehyde 10% in water solution, is used as a disinfectant and to preserve biological specimens. Formaldehyde 10% Buffered is highly suited for high-throughput laboratories owing to its brief fixing time, but it can also be utilized for long-term tissue storage.

What did they replace formaldehyde with? ›

Ethyl alcohol/polyethylene glycol, glutaraldehyde and phenoxyethanol are alternatives to formaldehyde, although these may pose other worker health and safety concerns.

How to make 4% formaldehyde? ›

For 1 L of 4% Formaldehyde, add 800 mL of 1X PBS to a glass beaker on a stir plate in a ventilated hood. Heat while stirring to approximately 60 °C. Take care that the solution does not boil. Add 40 g of paraformaldehyde powder to the heated PBS solution.

Can I use formaldehyde instead of paraformaldehyde? ›

Paraformaldehyde is a polymer of formaldehyde. Paraformaldehyde itself is not a fixing agent, and needs to be broken down into its basic building block formaldehyde. This can be done by heating or basic conditions until it becomes solubilized. Once that occurs, essentially they are exactly the same.

How do you calculate formaldehyde solution? ›

By using C1V1=C2V2, where C1 is 40% formaldehyde, V1 is volume of 40% formaldehyde, C2 is 0.001% formaldehyde, find your V2 (volume of 0.001% formaldehyde). V2=C1V1/C2. A log serial dilution will be of help to avoid obtaining a large V2. For making 10 ppm solution you need to perform dilution.

What should you not mix formaldehyde with? ›

Formaldehyde reacts with hydrochloric acid to form the potent carcinogen, bis-chloromethyl ether. Formaldehyde reacts with nitrogen dioxide, nitromethane, perchloric acid and aniline, or peroxyformic acid to yield explosive compounds. A violent reaction occurs when formaldehyde is mixed with strong oxidizers.
